A steamboat was stranded in the Mississippi River, and the Captain could not get her free. Eventually a hard-looking fellow came on board and said: "Captain, I understand you want a pilot to take you out of this difficulty?"

The Captain said, "Are you a pilot?"

"Well they call me one."

"Do you know where the snags and sandbars are" the Captain asked.

"No, sir."

"Well how do you expect to take me out of here if you don't know where the snags and sandbars are?"

"Because I knows where they ain't!" was his reply.

They arn't any such thing as Snags and sandbars.

By saying he doesn't know where the snags and sandbars are, the Pilot implies that he has never found a snag or sandbar by hitting one.

By saying he knows where the snags and sandbars aren't, he implies that he knows how to navigate the waterway without hitting them.

I'm sure everyone knows that he is slyly ridiculing the Captain. I suppose that is the joke.
